AP2M1 Antibody (YA3106) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to AP2M1.

Background

AP2M1 is a component of the adaptor protein complex 2 (AP-2). AP-2 functions in protein transport via transport vesicles across different membrane traffic pathways, acting as vesicle coat components involved in cargo selection and vesicle formation. AP-2 participates in clathrin-dependent endocytosis, incorporating cargo proteins into clathrin-coated vesicles (CCVs) destined for fusion with the early endosome. The clathrin lattice serves as a mechanical scaffold but cannot directly bind membrane components, whereas AP complexes interact with both the clathrin lattice and membrane lipids/proteins, serving as major clathrin adaptors for CCV formation. AP-2 also acts as a cargo receptor to selectively sort membrane proteins in receptor-mediated endocytosis and plays a role in recycling synaptic vesicle membranes from the presynaptic surface. AP-2 recognizes Y-X-X-[FILMV] (Y-X-X-Phi) and [ED]-X-X-X-L-[LI] endocytosis signal motifs (by similarity) and may regulate post-endocytic trafficking via the ARF6-dependent non-clathrin pathway. During hippocampal long-term potentiation, AP-2 mediates ADAM10 endocytosis. The AP-2 mu subunit binds transmembrane cargo proteins, recognizing Y-X-X-Phi motifs (by similarity), which become accessible upon Thr-156 phosphorylation in membrane-associated AP-2. This phosphorylation event involves assembled clathrin, which activates the AP-2 mu kinase AAK1. AP2M1 also contributes to Wnt-induced endocytosis of frizzled family members (by similarity).

  • Western blot analysis was performed on extracts from Hela (lane 1, 20 μg), 293(lane 2, 20 μg) using AP2M1 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Mouse/Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
