ANXA10 (annexin A10) is a member of the annexin family, primarily mediating biological effects through calcium-dependent phospholipid-binding and calcium ion binding. Gene chip analysis of L5 spinal cord after spinal nerve ligation (SNL)-induced neuropathic pain in mice revealed Anxa10 mRNA as the most upregulated gene in the annexin family (73.7-fold increase). While previous studies implicate several annexins in nociceptive pain, Anxa10's role remained undefined. SNL persistently upregulated Anxa10 mRNA and protein in mouse spinal cord, with ANXA10 colocalizing with neuronal marker MAP2 and astrocytic marker GFAP but not microglial marker CD11b. Pretreatment with Anxa10 siRNA partially prevented SNL-induced mechanical allodynia and heat hyperalgesia, while posttreatment attenuated neuropathic pain, suggesting ANXA10 as a novel therapeutic target. Anxa10 is the most upregulated annexin gene in neuropathic pain conditions and contributes to pain development/maintenance in mice.
