Angiopoietin-like 3 (ANGPTL3) is a hepatokine that regulates lipid and glucose metabolism by inhibiting lipoprotein lipase (LPL) activity through indirect mechanisms involving proprotein convertases PCSK6 and FURIN, thereby increasing plasma triglyceride (TG) levels. It also inhibits endothelial lipase to elevate HDL cholesterol and phospholipids while promoting adipocyte lipolysis. ANGPTL3 modulates VLDL-TG distribution to white adipose tissue (WAT) in response to feeding, potentially cooperating with ANGPTL8 and ANGPTL4. Additionally, it binds integrin αVβ3 on endothelial cells, activating FAK/MAPK/Akt signaling to promote angiogenesis, cell adhesion, and migration. In podocytes, it may influence glomerular endothelial cell properties and motility via integrin αVβ3 and Rac1 activation. ANGPTL3 also regulates hematopoietic stem cell (HSC) activity, possibly through IKZF1/IKAROS downregulation.

  Western blot analysis of extracts from 293 (lane 2(20μg)) and Mouse liver (lane 3(20μg)) using ANGPTL3 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (beta Actin, HY-P83730, 1/5000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit/Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
