ALKBH1 Antibody (YA628) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ALKBH1.

Background

ALKBH1 is a dioxygenase that acts on nucleic acids, such as DNA and tRNA, requiring molecular oxygen, alpha-ketoglutarate, and iron. It mainly acts on tRNAs, mediating their demethylation or oxidation depending on the context and subcellular compartment. ALKBH1 primarily functions as a tRNA demethylase, removing N(1)-methyladenine from various tRNAs, with a preference for N(1)-methyladenine at position 58 (m1A58) on tRNA stem loop structures. It regulates translation initiation and elongation in response to glucose deprivation by demethylating tRNA(Met) and preferentially recruiting N(1)-methyladenine-containing tRNAs to polysomes. In mitochondria, ALKBH1 interacts specifically with mt-tRNA(Met), mediating the oxidation of methylated cytosine(34) to form 5-formylcytosine (f(5)c), which enables recognition of the AUA codon in addition to AUG, expanding codon recognition in mitochondrial translation. ALKBH1 also demethylates DNA methylated on the 6th position of adenine (N(6)-methyladenosine) DNA, present at some L1 elements in embryonic stem cells, likely promoting their silencing. It demethylates mRNAs containing N(3)-methylcytidine modification and can repair alkylated single-stranded DNA by oxidative demethylation, albeit with low activity. ALKBH1 also exhibits DNA lyase activity, introducing double-stranded breaks at abasic sites, cleaving both single-stranded and double-stranded DNA, with the greatest activity towards double-stranded DNA with two abasic sites. This DNA lyase activity does not require alpha-ketoglutarate and iron and forms an irreversible covalent protein-DNA adduct with the 5’ DNA product. This activity is not required during base excision repair and class switch recombination of the immunoglobulin heavy chain during B lymphocyte activation. ALKBH1 may play a role in placental trophoblast lineage differentiation.
