Background	ALK-1 is a type I receptor for TGF-beta family ligands BMP9/GDF2 and BMP10 and plays a crucial role in regulating normal blood vessel development. Upon ligand binding, it forms a receptor complex with two type II receptors, where the type II receptors phosphorylate and activate ALK-1, leading to its autophosphorylation and subsequent binding and activation of SMAD transcriptional regulators.
