CD156B is a transmembrane metalloprotease that mediates the ectodomain shedding of numerous transmembrane proteins, including adhesion proteins, growth factor precursors, and cytokines critical for inflammation and immunity. It cleaves the membrane-bound precursor of TNF-alpha into its mature soluble form and facilitates the proteolytic release of soluble JAM3 from endothelial cell surfaces. Additionally, CD156B is responsible for shedding other cell-surface proteins such as p75 TNF receptor, interleukin-1 receptor type II, p55 TNF receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1, and amyloid precursor protein. It activates the Notch pathway by cleaving Notch to generate the membrane-associated intermediate fragment NEXT and plays a role in the proteolytic processing of ACE2. In hemostasis, CD156B sheds GP1BA (the platelet glycoprotein Ib alpha chain) (by similarity). It also mediates the proteolytic cleavage of LAG3 and IL6R, releasing their soluble forms (LAG3 by similarity; IL6R supported by references). Upon NK cell stimulation, CD156B cleaves FCGR3A, enhancing NK cell motility and detachment from opsonized targets. Furthermore, it cleaves TREM2, resulting in ectodomain shedding.

  Western blot analysis was performed on extracts from Hela (lane 1, 20 μg), 293 (lane 2, 20 μg), and Jurkat (lane 3, 20 μg) using ADAM17 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
