Background

Activin Receptor Type IA (ACVR1) is a bone morphogenetic protein (BMP) type I receptor involved in diverse biological processes including bone, heart, cartilage, nervous system and reproductive system development. Forming heterotetrameric complexes with type II receptors (AMHR2, ACVR2A or ACVR2B), it becomes transphosphorylated upon ligand (e.g., BMP7 or GDF2/BMP9) binding, subsequently phosphorylating SMAD1/5/8 to mediate BMP signaling (by similar: may activate non-canonical p38 MAPK pathways). ACVR1 also suppresses TGFβ/activin pathway signaling by competitively inhibiting activin binding to type II receptors (by similar: potentially promotes HAMP expression through BMP6 interaction).
