Background

Actin Related Protein 2 (Arp2) serves as the essential ATP-binding component of the Arp2/3 complex, mediating actin polymerization upon activation by nucleation-promoting factor (NPF) (by similar). It drives cell motility by facilitating the formation of branched actin networks in the cytoplasm and may participate in recognizing the pointed ends of daughter actin filaments. In podocytes, Arp2 is regulated by AVIL and PLCE1 to enable lamellipodia formation. Remarkably, the Arp2/3 complex also functions in the nucleus, where it regulates gene expression and DNA damage repair through nuclear actin polymerization (by similar), playing a critical role in homologous recombination (HR) repair of DNA double-strand breaks (DSBs).
