all-trans-Anhydro Retinol (Anhydrovitamin A) is an orally active degradation product formed by the dehydration of vitamin A alcohol and its esters under conditions such as acidic catalysis or hydroxylic solvents, with a biological activity only 0.4% of that of Vitamin A (HY-B1342). In rats, all-trans-Anhydro Retinol can be metabolized into hydroxy derivatives and their esters, but it cannot be converted into Vitamin A (HY-B1342) and has a low storage capacity in the body[1][2].

In Vivo

all-trans-Anhydro Retinol (2.22 mg; oral administration/subcutaneous injection; 6 days) in Vitamin A (HY-B1342)-deficient rats shows that it is metabolized into various monohydroxy, dihydroxy and ester derivatives through hydroxylation and esterification, with no detectable Vitamin A (HY-B1342) or unmetabolized Anhydrovitamin A in the liver[1].

Animal Model: Male Wistar rats (approximately 7 weeks old) made Vitamin A (HY-B1342)-deficient by dietary restriction[1]
Dosage: 0.22 mg/0.1 mL
Administration: Oral administration/subcutaneous injection; 6 days
Result: Was metabolized into at least seven derivatives in the liver, including non-hydroxy, monohydroxy, and dihydroxy compounds with ester groups. The metabolites were poorly absorbed and stored, with no detectable levels remaining in the liver after 30 days.
Resulted in 26% of the liver storage by subcutaneous injection compared to oral administration. The growth-promoting activity was attributed to the in vivo-formed derivatives rather than conversion to vitamin A.
