Description

AG-7404 is an orally active, irreversible inhibitor of the picornavirus 3C protease. AG-7404 blocks the processing of viral polyproteins, thereby inhibiting viral replication. AG-7404 has synergistic antiviral activity with capsid inhibitors such as V-073 (HY-104074) or BTA798 (HY-106254) and is effective against V-073-resistant variants. AG-7404 is used in the study of enterovirus infections such as poliovirus[1][2][3].

In Vitro

AG-7404 (0.080-0.674 μM; 3 d) inhibits viral replication in HeLa cells with poliovirus with an EC50 of 0.080-0.674 μM[1].
AG-7404 (0.218-0.819 μM; 3 d) remains active in V-073-resistant poliovirus strains with an EC50 of 0.218-0.819 μM[1].

Cell Viability Assay[1]

Cell Line: HeLa cells (for wild-type and vaccine-derived poliovirus strains)
Concentration: 0.080-0.674 μM
Incubation Time: 3 days
Result: Dose-dependently reduced viral cytopathic effect (CPE) as assessed by crystal violet staining and CTG assay.
EC50 values were determined for each strain, demonstrating broad activity against wild-type, circulating vaccine-derived (cVDPV), and immunodeficient vaccine-derived (iVDPV) strains.
