ABT-255 free base is an orally active anti-bacterial agent. In Vitro

ABT-255 (0.016-0.5 μg/mL) free base shows MIC values of 0.016 μg/mL against ATCC 35801, 0.031 μg/mL against ATCC 25618, and 0.031 μg/mL against both 35837b (Ethambutol (HY-B0535)-resistant) and 35838c (Rifampin (HY-B0272)-resistant)[1].

In Vivo

ABT-255 (3.13-25 mg/kg, p.o., once daily, 4 weeks) free base shows efficacy against S. aureus, S. pneumoniae and E. coli and reduces viable counts of drug-resistant Mycobacterium tuberculosis in CF-1 mice[1].

Animal Model: 4- to 6-week-old female CF-1 mice were intraperitoneally injected with overnight cultures of Staphylococcus aureus NTCC 10649, Streptococcus pneumoniae 6303, or Escherichia coli JUHL, with the inoculum adjusted to approximately 100 times the 50% lethal dose[1]
Dosage: 3.13, 6.25, 12.5, 25 mg/kg
Administration: p.o. once daily for 4 weeks
Result: Reduced Staphylococcus aureus NTCC 10649, Streptococcus pneumoniae 6303, or Escherichia coli in mice JUHL.
Animal Model: 4- to 6-week-old female CF-1 mice were intravenously injected via the caudal tail vein with approximately 107 viable M. tuberculosis cells suspended in 0.2 mL of modified 7H10 broth[1]
Dosage: 6.25, 12.5, 25 mg/kg
Administration: p.o. once daily for 4 weeks
Result: Produced a dose-responsive 0- to 5.5-log reduction in M. tuberculosis (ATCC 35801) counts in lung tissue.
Produced a dose-related 1- to 3-log reduction in viable Ethambutol (HY-B0535)-resistant M. tuberculosis (ATCC 35837) counts in lung tissue.
Produced a dose-responsive 1- to 2.5-log reduction in viable Rifampin (HY-B0272)-resistant Mycobacterium tuberculosis (ATCC 35838) counts in lung tissue.
