8pyDTZ is a pyridyl diphenylterazine (DTZ) analog and an ATP-independent pyridyl substrate of LumiLuc luciferase. 8pyDTZ exhibits spectrally shifted emission. 8pyDTZ has excellent biocompatibility and superior in vivo sensitivity. 8pyDTZ can be used for in vivo luminescence imaging[1].

In Vitro

Guidelines (The following is our recommended protocol. This protocol is provided as a guide only and should be modified to suit your specific needs).
1 Working Solution Preparation [2]
1) Prepare a solution of 8pyDTZ in an aqueous injectable formulation containing 10% Ethanol (v/v), 10% Glycerol (HY-B1659) (v/v), 10% 2-Hydroxypropyl-β-cyclodextrin (HPBCD, w/v), and 35% PEG-300 (HY-Y0873) (v/v).
2) A typical working solution is 4 μmol (1.5 mg) 8pyDTZ in 480 μL of formulation.
Note: This product is unstable in solution. It is recommended that you prepare the solution immediately before use.
2 Specific staining steps
1) Anesthetize the mouse by isoflurane inhalation for 5 min, remove the hair in the imaging area with an electric shaver, apply Nair depilatory cream for 1 min, wipe the remaining hair with sterile gauze, rinse with DPBS and disinfect with 70% ethanol.
2) Inject 8pyDTZ working solution intraperitoneally into the left lower abdomen of the mouse.
3) Immediately place the mouse in a bioluminescence imaging system (such as the Ami HT system) and set the parameters: emission filter fully open, field of view 25 cm, aperture f-stop 1.2, merge mode 1×1, exposure time 1 s.
4) Collect bioluminescence images every 1 min for 20 min. Use Fiji (ImageJ) software to analyze the images, and use Excel for data processing.

In Vivo

LumiLuc-8pyDTZ pair is compared with several benchmark reporters in a tumor xenograft mouse model. LumiLuc-8pyDTZ shows high sensitivity and does not need organic cosolvents for in vivo administration. LumiScarlet-8pyDTZ is comparable to Akaluc-AkaLumine, the brightest ATP-dependent luciferase-luciferin pair, for detecting cells in deep tissues of mice[1].
