5-LOX-IN-8 is a 5-LOX inhibitor with anti-inflammatory activity. 5-LOX-IN-8 suppresses IL-6, IL-1β, TNF-α and IFN-γ in macrophages and reduces IL-8 secretion in SW480 cells. 5-LOX-IN-8 reduces disease activity in a DSS colitis model. 5-LOX-IN-8 can be used for research of inflammatory bowel disease (IBD)[1].

In Vitro

5-LOX-IN-8 (Compound 18d) (0.3-50 μM, 24 h) shows cytotoxicity in THP-1 cell (IC50: 37.65 μM) and SW 480 cell (IC50: 14.37 μM)[1].
5-LOX-IN-8 (10 μM, 5 min) inhibits human recombinant 5-LOX by 40.65%, and also inhibits NO production by 38.85% in SW480 cells[1].
5-LOX-IN-8 (10 μM, 1 h pretreatment and 24 h co-culture with LPS) suppresses IL-6 (93.98%), IL-1β (19.41%), TNF-α (45.97%) and IFN-γ (66.61%) production in LPS (HY-D1056)-stimulated THP-1 macrophages[1].

Cell Viability Assay[1]

Cell Line: THP-1 cell, SW 480 cell
Concentration: 0.3-50 μM
Incubation Time: 24 h
Result: Inhibited the cell viability of THP-1 cell (IC50: 37.65 μM) and SW 480 cell (IC50: 14.37 μM).
And the cell viability of THP-1 cell and SW 480 cell at 50 μM were 65.54% and 56.23%, respectively.
In Vivo

5-LOX-IN-8 (Compound 18d) (30 mg/kg) reduces disease activity in 5% DSS (HY-116282C) induced murine colitis model[1].

Dosage: 30 mg/kg
Administration:
Result: Increased the body weight (87.84%) compared with DSS-treated group (73.75%).
Slightly increased the colon length (mm) compared with DSS-treated group.
Significantly reduced DAI from Day-4 to Day-7, with a Day-7 DAI of 8.67 compared to 11.67 for the DSS-treated group.
