Description

5-AF594 tyramide is a red fluorescent dye (Ex=590 nm, Em=618 nm). 5-AF594 tyramide is utilized as reporter fluorescent substrate of horseradish peroxidase (HRP)-catalyzed deposition for tyramide signal amplification (TSA). 5-AF594 tyramide can be used for multiplex immunohistochemistry (mIHC)[1].

In Vitro

5-AF594 tyramide assay
a. Slides are permeabilized in 0.25% Triton X-100 10 min at RT.
b. Incubate overnight at 4 °C in the primary antibody.
c. Incubate in 3% H2O2 in 1x PBS for 20 min to block endogenous peroxidase activity.
d. Detect primary antibody with anti-rabbit IgG conjugated to FITC for 1 h at RT (1: 100), and then apply an anti-FITC-conjugated HRP antibody (1: 500) overnight at 4 °C.
e. Apply a 5-AF594 tyramide for 10 min (1:200).
f. Fluorescence microscopy detection.
Note: it is suggested that the optimal concentration of 5-FITC tyramide working solution according to actual conditions.
