NC762 is a humanized IgG1κ monoclonal antibody that targets human B7-H4 (B7 homolog 4). NC762 has enhanced binding to CD16a (FcγRIIIa) after being Fc engineered with three point mutations (S239D/A330L/I332E; DLE) and demonstrate increased antibody-dependent cellular cytotoxicity (ADCC) activity. NC762 inhibits tumor growth in vivo through binding to tumor-expressing B7-H4. NC762 can be studies in research on cancer such as advanced or metastatic solid tumors.

NC762 (1-30 mg/kg, i.p., Q7D for 4 doses) reduces tumor volume dose-dependently in NSG mice subcutaneous inoculated with 1E06 624Mel.hB7H4 cells and 3E05 human PBMCs[2].
NC762 (10 mg/kg, i.p., Q4D for 7 doses) reduces anti-B7H4 mediated tumor growth with NK cell contribution and extends survival in NSG mice model subcutaneous inoculated with 1E06 624Mel.hB7H4 cells mixed with either 1E06 total or NK-cell depleted PBMCs[2].
NC762 (10 mg/kg, i.p., Q7D for 4 doses) restricts tumor growth in NSG mice model subcutaneous inoculated with 1E06 624Mel.hB7H4 cells in either absence of PBMCs or the presence of human T cells[2].
NC762 (10 mg/kg, i.p., Q7D for 4 doses) inhibits tumor growth with similar activity as a mutant of NC762 that has a low Fc affinity in NSG mice subcutaneous inoculated with 1E06 624Mel.hB7H4 cells, suggesting an ADCC-independent mechanism of tumor growth restriction[2].
