eGFP mRNA-LNP (500 ng/μL) is a lipid nanoparticle (LNP) containing eGFP mRNA. eGFP mRNA-LNP carries Enhanced Green Fluorescent Protein (eGFP), which will express green fluorescent protein after entering the cell. eGFP is commonly used as a reporter gene detectable by fluorescence microscopy or flow cytometry (excitation wavelength = 488 nm; emission wavelength = 535/40 bandpass). eGFP mRNA-LNP can be used for RNA delivery, translation efficiency and cell viability[1].

In Vitro

eGFP mRNA-LNP (500 ng/μL) (1 μM, 24 h) results in full mRNA formulation translation into eGFP protein at significant amounts in HeLa cell line, whereas the translation by macrophages is low[2].
eGFP mRNA-LNP (1 μM, 24 h) results in weak signals in Kupffer cells (KC) , whereas hepatocytes expresses eGFP in visible amounts throughout the whole cell[2].
eGFP mRNA-LNP (0.25-1 μM, 24-48 h) shows time-dependent eGFP transfection at 0.25 μM, and time does not make much of a difference at 1 μM in alveolar epithelial cells and human primary lung fibroblasts[3].
eGFP mRNA-LNP (0.4 μM, 48 h) shows over half of the cell lines exhibit medium or poor transfection whilst human colon epithelial HCT116 and human hepatocyte HuH-7 cells are the high expressors[4].

Immunofluorescence[2]

In Vivo

eGFP mRNA-LNP (500 ng/μL) (2 mg/kg, i.v., single dose) allows detection of eGFP signals only in hepatocytes of C57Bl/6J mice through intravital imaging 16 h after eGFP mRNA-LNP administration[2].
eGFP mRNA-LNP (500 ng/μL) (3 mg/kg, i.v., single dose) allows significant and detectable eGFP protein expression level in Kupffer cells (KC) and monocytic macrophages (MoMϕ) in C57Bl/6J[2].
