Background

PI3 Kinase p110 beta is a phosphoinositide-3-kinase (PI3K) that phosphorylates phosphatidylinositol derivatives at the 3-position of the inositol ring to produce 3-phosphoinositides. It utilizes ATP and PtdIns(4,5)P2 (phosphatidylinositol 4,5-bisphosphate) to generate phosphatidylinositol 3,4,5-trisphosphate (PIP3), which recruits PH domain-containing proteins like AKT1 and PDPK1 to the membrane, activating signaling cascades involved in cell growth, survival, proliferation, motility, and morphology. This kinase participates in AKT1 activation upon stimulation by G-protein coupled receptor (GPCR) ligands such as CXCL12, sphingosine 1-phosphate, and lysophosphatidic acid, and may act downstream of receptor tyrosine kinases. It is essential for stable platelet adhesion and aggregation in various signaling pathways, regulating platelet activation triggered by GPCRs, α-IIb/β3 integrins (ITGA2B/ITGB3), and ITAM (immunoreceptor tyrosine-based activation motif)-bearing receptors like GP6. It also modulates adhesion strength of ITGA2B/ITGB3-activated receptors for cellular force transmission and is required for platelet aggregation induced by F2 (thrombin) and thromboxane A2 (TXA2). The protein plays roles in cell survival, migration, early autophagosome formation, and PtdIns3P (phosphatidylinositol 3-phosphate) level modulation, potentially acting as a scaffold to positively regulate autophagy independently of its lipid kinase activity. In insulin signaling, it may function as a kinase-independent scaffold and regulate cell proliferation and clathrin-mediated endocytosis. It mediates oncogenic signals in PTEN-deficient cell lines, where its lipid kinase activity is critical for transformation, and is necessary for ERBB2- and RAS-driven tumor growth. Additionally, it exhibits protein kinase activity capable of autophosphorylation.
