IGF-1R is a receptor tyrosine kinase that mediates the actions of insulin-like growth factor 1 (IGF1), binding IGF1 with high affinity and IGF2/insulin (INS) with lower affinity. Activated IGF1R controls cell growth and survival, playing a critical role in tumor transformation and malignant cell survival. Ligand binding triggers receptor autophosphorylation and tyrosine phosphorylation of substrates like IRS1/2, Shc, and 14-3-3 proteins, activating the PI3K-AKT and Ras-MAPK pathways. MAPK activation promotes proliferation, while PI3K activation inhibits apoptosis and stimulates protein synthesis via AKT/mTOR signaling and BAD inactivation. Phosphorylated IRS1 also recruits Grb2/SOS to activate Ras-MAPK. Additionally, IGF1R signals through JAK/STAT, with STAT3 activation potentially essential for its transforming activity. IGF1 inhibits JNK via MAP3K5/ASK1 phosphorylation. In hybrid receptors with INSR, IGF1R binds IGF1. by similar studies show that IGF1R-INSR Long hybrids are activated by IGF1 (high affinity) and IGF2 (low affinity) but not insulin, while IGF1R-INSR Short hybrids respond to IGF1, IGF2, and insulin. However, other research suggests both hybrid types share similar binding characteristics, primarily recognizing IGF1 with low insulin affinity.
