FMR1 is a multifunctional polyribosome-associated RNA-binding protein that plays a central role in neuronal development and synaptic plasticity by regulating alternative mRNA splicing, stability, dendritic transport, and local protein synthesis. It mediates liquid-liquid phase separation to assemble mRNAs into cytoplasmic ribonucleoprotein granules, concentrating mRNAs with regulatory factors. FMR1 stabilizes DLG4/PSD-95 and MBP mRNAs in hippocampal neurons and glial cells, respectively, with enhanced stabilization upon mGluR stimulation (by similar). It selectively delivers dendritic mRNAs to synapses (by similar) and forms translation-repressive granules via phosphorylation and CAPRIN1 interaction. FMR1 acts as a translational repressor by stalling ribosomes (by similar) and participates in miRNA-mediated repression, while also activating translation of synaptic mRNAs (by similar). It binds numerous dendritic mRNAs, recognizing consensus sequences (e.g., 5'-ACU[GU]-3') and G-quadruplex structures. FMR1 facilitates nuclear export of m6A-modified mRNAs via XPO1/CRM1 and modulates ion channel activity (e.g., KCNT1, CACNA1B) and microtubule dynamics (by similar). It may influence DNA damage response (DDR) and positively regulates influenza A virus (IAV) replication by assisting viral ribonucleoprotein (vRNP) assembly and export.
