Background

Cytochrome P450 4A is a cytochrome P450 monooxygenase involved in the metabolism of fatty acids and their oxygenated derivatives (oxylipins). Mechanistically, it uses molecular oxygen to insert one oxygen atom into a substrate and reduces the second into water, with electrons provided by NADPH via cytochrome P450 reductase (CPR; NADPH-ferrihemoprotein reductase). It predominantly catalyzes the omega-oxidation of saturated and unsaturated fatty acids, with catalytic efficiency decreasing in the order: dodecanoic > tetradecanoic > (9Z)-octadecenoic > (9Z,12Z)-octadecadienoic > hexadecanoic acid. It acts as the major omega-hydroxylase for lauric acid in the liver and participates in the omega-hydroxylation of arachidonate to produce 20-HETE, a signaling molecule with vasoconstrictive and natriuretic effects that influence arterial blood pressure. It can also perform omega-1 oxidation of fatty acids with lower efficiency and may contribute to the degradation of very long-chain fatty acids (VLCFAs) by initiating chain shortening through successive omega-oxidations. Additionally, it omega-hydroxylates the (9R,10S)-epoxy-octadecanoate stereoisomer but exhibits minimal activity toward long-chain 3-hydroxy fatty acids and prostaglandins A1 and E1.
