Leukotriene A4 methyl ester (LTA4 methyl ester) is synthesized in mast cells, eosinophils, and neutrophils from arachidonic acid by 5-lipoxygenase (5-LO), which exhibits both lipoxygenase and LTA4 synthase activities. LTA4 is rapidly metabolized by LTA4 hydrolase or LTC4 synthase to LTB4 or LTC4, respectively. LTA4, from leukocytes, is known to undergo transcellular metabolism in platelets, erythrocytes, and endothelial cells. Further metabolism of LTA4 by 15-LO leads to lipoxin biosynthesis. LTA4 as a free acid is highly unstable. The methyl ester is stable and can be readily hydrolyzed to the free acid as needed.

Siphonaxanthin is an inhibitor of fibroblast growth factor receptor-1 (FGFR-1) signaling transduction in vascular endothelial cells. Siphonaxanthin inhibits the proliferation, migration, and tube formation of human umbilical vein endothelial cells (HUVECs), as well as the outgrowth of microvessels in the rat aortic ring. Siphonaxanthin can also inhibit the translocation of the high-affinity IgE receptor (FcεRI) to lipid rafts in mast cells, acting as an anti-inflammatory agent. Imidazolidinyl urea has broad-spectrum antibacterial activity, which mainly inhibits the reproduction of gram-negative bacteria and gram-positive bacteria, and restricts the growth of yeast and mold to a certain extent. Imidazolidinyl urea can induce non-histaminergic allergy by MRGPRX2 activation of mast cells .

Toluidine blue (ZnCl2) is a basic thiazine dye commonly used as a biological stain for microscopy. It has a deep bluish-purple color and is commonly used to stain nucleic acids such as DNA and RNA, as well as to stain mast cells, cartilage, and other connective tissues. Toluidine blue (ZnCl2) stains the acidic components of these tissues, such as sulfated or carboxylated mucopolysaccharides. It is frequently used in histology, cytology, and pathology applications to aid in the diagnosis of various diseases and conditions. The dye is usually applied to tissue sections prior to microscopic examination and can be differentiated using an acidic alcohol solution. Toluidine blue (ZnCl2) is a relatively simple and inexpensive stain with good reproducibility, making it a popular choice for many laboratories.

SB 201146 is an LTB4 receptor antagonist with a potency range of 10 pM to 1 μM that plays a critical role in asthma-associated airway inflammation. Studies have focused on its effects on eosinophil survival, a hallmark of asthma pathology. Studies have shown that eosinophil-derived cysteinyl leukotrienes, including LTC4 and LTD4, as well as factors such as GM-CSF and fibronectin promote eosinophil survival. SB 201146 effectively reversed mast cell- and lymphocyte-induced eosinophil survival, highlighting its potential therapeutic role in disrupting the autocrine cysteinyl leukotriene pathway that maintains eosinophil viability. This antagonist also highlights the importance of LTB4 as a paracrine mediator that influences eosinophil survival in inflammatory settings such as asthma .

Z-3578 is a small-molecule antagonist targeting MrgX2 (Mas-related G protein-coupled receptor X2) with significant anti-pseudoallergic activity and a KD value of 729 nM. Z-3578 effectively inhibits mast cell degranulation induced by substance P (SP) and C48/80, suppressing the release of β-hexosaminidase with IC50 values of 4.90 µM and 6.18 µM, respectively. It also markedly reduces the release of histamine and TNF-α, along with intracellular calcium flux. In a murine pseudoallergy model, Z-3578 significantly alleviates paw swelling and dye extravasation and lowers serum histamine levels, indicating potent in vivo anti-allergic effects. Z-3578 holds promise as a lead compound for the treatment of allergic diseases, especially pseudoallergic reactions .
